Cell-Penetrating Peptides for Medical Theranostics and Targeted Drug Delivery
Abstract
We focus on three major topics in this chapter. First, we describe cellular uptake mechanisms of cell-penetrating peptides (CPPs). The pitfalls and complementary tools in conducting mechanistic studies of uptake are illustrated. Second, we depict applications of two powerful CPPs, activatable cell-penetrating peptides (ACPPs) and tumor internalizing RGD (iRGD) peptides, for medical imaging and drug delivery. A combinational approach with CPPs and the clustered regularly interspaced short palindromic repeats (CRISPR)- associated (Cas) protein 9 (CRISPR-Cas9) system may provide an alternative for gene delivery and editing. Lastly, we review the safety and biocompatibility of CPPs in medical applications.
