Determination of Indole-3-acetic Acid in Rieger Begonia Leaf Tissue by HPLC-fluorometry and GC-MS
Abstract
The paper reports on a study that deals with development of a high-performance liquid chromatography (HPLC) method for determination of Indole Acetic Acid (IAA) in Rieger Begonia (Begonia x hiemalis) leaf tissue. The method is based on reverse phase separation on C//1//8 column followed by fluorometric determination. Prior to analysis extensive sample preparation in form of acid-base partitioning and Sephadex column clean-up was found to be essential. The minimum detectable limit of 25 pg was obtained for IAA. The linear range for the method was determined to be 1 multiplied by 10**4. The results obtained by the method compared favorably with more elaborate gas chromatography-mass spectrometry determinations.
